摘要: 目的:研究广藿香挥发油对感染后肠易激综合征(PI-IBS)模型大鼠结肠黏膜上皮细胞紧密连接蛋白ZO-1、Occludin表达的影响。方法:将大鼠随机分为正常对照组(蒸馏水)、模型组(蒸馏水)、马来酸曲美布汀片组(TMT,0.1 g/kg)和广藿香挥发油低、中、高剂量组[2、3、4 g(生药)/kg],每组8只。除正常对照组外,其余各组大鼠均采用结肠灌注乙酸法复制PI-IBS模型。造模成功后,每天ig给药1次,连续给药5 d。采用免疫组化法检测结肠黏膜中ZO-1、Occludin蛋白表达[以积分光密度(IOD)值反映]并分析其分布情况。结果:正常对照组大鼠ZO-1、Occludin蛋白均匀分布在肠上皮细胞的顶端,呈蜂窝状或呈点状聚集;模型组大鼠ZO-1、Occludin蛋白零散分布在肠上皮细胞的顶端,其染色分布不均或褪色;广藿香挥发油组大鼠ZO-1、Occludin蛋白分布类似正常对照组,但其染色浅于正常对照组。模型组大鼠ZO-1、Occludin蛋白的IOD值低于正常对照组(P<0.01);广藿香挥发油高剂量组大鼠ZO-1、Occludin蛋白的IOD值,以及广藿香挥发油中剂量组、TMT组大鼠ZO-1蛋白的IOD值高于模型组(P<0.05或P<0.01)。结论:广藿香挥发油可上调PI-IBS大鼠肠黏膜上皮细胞紧密连接蛋白ZO-1和Occludin的表达。
ABSTRACT: OBJECTIVE: To study the effects of volatile oil of Pogostemon cablin on the expression of tight junction proteins ZO-1 and Occludin in colonic mucosal epithelial cells of rats with post-infectious irritable bowel syndrome (PI-IBS). METHODS: Rats were randomly divided into normal control group (distilled water), model group (distilled water), Trimebutine maleate tablet group (TMT, 0.1 g/kg) and volatile oil of P. cablin low-dose, medium-dose and high-dose groups [2, 3, 4 g(crude drug)/kg] with 8 rats in each group. Except for normal control group, those groups were given colon perfusion of acetic acid to induce PI-IBS model. After modeling, they were given relevant medicine intragastrically once a day for consecutive 5 days. The expression of ZO-1 and Occludin [by IOD] and their distribution were detected by immunohistochemistry. RESULTS: In normal control group, ZO-1 and Occludin evenly distributed on the top of enterocyte, manifesting as alveolate and punctiform; in model group, ZO-1 and Occludin scattered on the top of enterocyte, showing uneven dyeing or fade; the distribution of ZO-1 and Occludin in volatile oil of P. cablin groups was similar to normal control group, and its dyeing was lighter than that of normal control group. IOD value of ZO-1 and Occludin in model group were lower than in normal control group (P<0.01); those of volatile oil of P. cablin high-dose group and the IOD value of ZO-1 of P. cablin medium-dose group and TMT group were higher than those of model group (P<0.05 or P<0.01). CONCLUSIONS: The volatile oil of P. cablin can up-regulate the expression of tight junction proteins ZO-1 and Occludin in colonic mucosal epithelial cells of rats with PI-IBS.
