摘要: 目的:优化微波协同双水相提取甘草中总黄酮的工艺。方法: 以硫酸铵和乙醇为双水相萃取体系,微波法提取甘草中的总黄酮;采用单因素试验和正交试验考察硫酸铵质量浓度、乙醇体积分数、固液比、微波功率、提取时间 5个因素对总黄酮提取率的影响,优化提取工艺并进行验证试验。结果:最优提取工艺为硫酸铵质量浓度0.40 g/ml、乙醇体积分数70%、固液比1 ∶ 25、提取时间3 min、微波功率300 W;验证试验中甘草总黄酮平均提取率为2.038%(RSD=0.69%,n=3),双水相萃取率为94.46%(RSD=1.30%,n=3)。结论:该工艺操作简单、结果稳定、反应条件温和、提取效率较高,适合工业化提取甘草中总黄酮。
ABSTRACT: OBJECTIVE: To optimize microwave-assisted aqueous two-phase system of total flavonoids from Glycyrrhiza uralensis. METHODS: The ammonium sulfate and ethanol were used as aqueous two-phase extraction system, the method of microwave was applied to extract total flavonoids from G. uralensis. The effects of 5 factors on extraction rate of total flavonoids were investigated by single factor test and orthogonal test, such as the mass concentration of ammonium sulfate, ethanol volume fraction, solid-liquid ratio, microwave power and extraction time. Validation test was conducted. RESULTS: The optimal extraction condition was as follows as the mass concentration of ammonium sulfate of 0.40 g/ml, ethanol volume fraction of 70%, solid-liquid ratio of 1 ∶ 25, extraction time of 3 min and microwave power of 300 W; in validation test, the extraction rate of total flavonoids from G. uralensis was 2.038% (RSD=0.69%, n=3), and the extraction rate of two-phase system was 94.46% (RSD=1.30%, n=3). CONCLUSIONS: The technology is simple in operation and stable in results, and also has gentle reaction conditions and high extraction rate, which is suitable for industrial production.
