摘要: 目的:优化荜茇总生物碱的提取工艺,为荜茇的深入开发和综合利用提供参考。方法:以总生物碱(指标性成分为胡椒碱)的提取率为考察指标,药材粉碎度、液料比、提取时间、提取次数为考察因素,采用单因素试验和Box-Behnken响应面法优化荜茇总生物碱的提取工艺,并进行3次(荜茇用量分别为1.00、10.00、100.00 g)验证试验。结果:最优提取工艺为粉碎度90目,加入32倍量的无水乙醇提取3次,每次提取170 min。最优工艺条件下,在荜茇用量为1.00、10.00、100.00 g时荜茇总生物碱的提取率分别为5.015、5.028、5.041 mg/g,与预测值5.025 mg/g的相对误差分别为0.20%、0.06%、0.26%,差异较小。结论:本研究优选的荜茇总生物碱的提取工艺合理、稳定、可行。
ABSTRACT: OBJECTIVE: To optimize the extraction technology of total alkaloids from Piper longum, and to provide reference for deep development and comprehensive utilization of P. longum. METHODS: With the extraction rates of total alkaloids (the target component for piperine) as reference index, using comminution degree of medicinal materials, liquid-solid ratio, extraction time and extraction times as factors, single factor test and Box-Behnken response surface methodology were adopted to optimize the extraction technology of total alkaloids from P. longum. The validation tests were conducted for 3 times (the amounts of P. longum were 1.00, 10.00, 100.00 g). RESULTS: The optimal extraction technology was that comminution degree of 90 mesh, 32-fold absolute ethanol, extracting for 3 times, lasting for 170 min each time. Under optimal technology, the extraction rates of total alkaloids from P. longum were 5.015, 5.028, 5.041 mg/g when the amount of P. longum were 1.00, 10.00, 100.00 g; relative errors of them to predicted value 5.025 mg/g were 0.20%, 0.06%, 0.26%, respectively. There was small difference. CONCLUSIONS: The optimal extraction technology of total alkaloids of P. longum is reasonable, stable and feasible.
