摘要: 目的:研究白耙齿菌固体发酵物对庆大霉素(GM)所致大鼠急性肾衰竭的改善作用。方法:将50只大鼠随机分为空白对照组、模型组、阳性对照组(尿毒清颗粒,2 g/kg)和白耙齿菌固体发酵物低、高剂量组(2、4 g/kg),每组10只。除空白对照组外,其余各组大鼠均连续7d腹腔注射GM(140 mg/kg)复制急性肾衰竭模型。给药组大鼠每天在造模2 h后灌胃给药1次,空白对照组和模型组在相应时间灌胃等量生理盐水;造模结束后,各组大鼠再连续给药7 d,每天给药1次。给药结束后,检测大鼠血肌酐(Scr)、血尿素氮(BUN)、尿肌酐(Ucr)和右肾组织中白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)含量,苏木精-伊红染色后观察大鼠左肾组织病理学变化。结果:与空白对照组比较,模型组大鼠Scr、BUN、Ucr和右肾组织中IL-6、TNF-α含量均显著增加(P<0.01);左肾组织发生肾小管上皮细胞明显水肿、变性、细胞核消失以及管腔内蛋白样沉积增多等病理学变化。与模型组比较,阳性对照组大鼠Ucr和白耙齿菌固体发酵物低剂量组大鼠BUN以及白耙齿菌固体发酵物高剂量组大鼠Scr、BUN、Ucr含量均显著减少(P<0.05或P<0.01);同时给药组大鼠右肾组织中IL-6、TNF-α含量均显著减少(P<0.05或P<0.01);左肾组织的炎症浸润减轻、肾小管和血管的增生受到抑制,病理学变化得到明显改善。结论:白耙齿菌固体发酵物对GM所致急性肾衰竭具有一定的改善作用。
ABSTRACT: OBJECTIVE: To study the improvement effects of Irpex lacteus solid fermentation products on gentamycin (GM)-induced acute renal failure in rats. METHODS: Fifty rats were randomly divided into blank control group, model group, positive control group (Niaoduqing granule, 2 g/kg) and Irpex lacteus solid fermentation products low-dose and high-dose groups (2, 4 g/kg), with 10 rats in each group. Except for blank control group, other groups were given GM (140 mg/kg) intraperitoneally for consecutive 7 d to induce acute renal failure model. Administration group was given relevant medicine intragastrically 2 h after modeling, once a day. Blank control group and model group were given constant volume of normal saline at corresponding time. After modeling, each group was given relevant medicine for consecutive 7 d, once a day. After medication, the contents of Scr, BUN, Ucr and IL-6, TNF-α in right renal tissue were detected. The pathological changes of left renal tissue was observed by HE staining. RESULTS: Compared with blank control group, the contents of Scr, BUN, Ucr and IL-6, TNF-α in right renal tissue were increased significantly in model group (P<0.01). The pathological changes of left renal tissue were observed, such as obvious renal tubular epithelial cell edema, degeneration, nucleus disappearance and the increase of protein-like deposition in the lumen. Compared with model group, the content of Ucr in positive control group and the content of BUN in I. lacteus solid fermentation products low-dose group and the contents of Scr, BUN, Ucr in I. lacteus solid fermentation products high-dose group were decreased significantly (P<0.05 or P<0.01); meanwhile, the contents of IL-6 and TNF-α in right renal tissue were decreased significantly in administration groups (P<0.05 or P<0.01). The inflammatory infiltration of left renal tissue was relieved, and renal tubules and vessels hyperplasia were inhibited; the pathological changes were improved significantly. CONCLUSIONS: I. lacteus solid fermentation products could improve GM-induced acute renal failure to certain extent.
