补肾活血化痰方对多囊卵巢综合征模型大鼠的治疗作用机制研究

| 摘要： | 目的：考察补肾活血化痰方对多囊卵巢综合征（PCOS）模型大鼠的治疗作用机制。方法：将50只大鼠随机分为正常对照组、模型对照组、阳性对照组[罗格列酮，3.0 mg/（kg·d）]和补肾活血化痰方高、低剂量组[7.5、5.0 g/（kg·d）]，每组10只。除正常对照组外，其余各组大鼠均采用Poresky法复制PCOS模型。造模后，正常对照组和模型对照组大鼠ig生理盐水，各给药组大鼠ig相应药液，每天1次，连续22 d。给药结束后，测定大鼠血清中睾酮（T）、抗苗勒氏管激素（AMH）、胰岛素样生长因子Ⅰ（IGF-Ⅰ）、肿瘤坏死因子α（TNF-α）水平和卵巢组织中转化生长因子β1（TGF-β1）阳性表达情况。结果：与正常对照组比较，模型对照组大鼠血清中T、AMH、IGF-Ⅰ、TNF-α水平及卵巢组织中TGF-β1阳性表达率均显著升高（P＜0.05）；与模型对照组比较，各给药组大鼠血清中T、AMH、IGF-Ⅰ、TNF-α水平及卵巢组织中TGF-β1阳性表达率均显著降低（P＜0.05），且补肾活血化痰方各剂量组指标优于阳性对照组（P＜0.05）。结论：补肾活血化痰方能明显降低PCOS模型大鼠血清中T、AMH、IGF-Ⅰ、TNF-α水平以及卵巢组织中TGF-β1阳性表达率，这可能是其治疗PCOS的作用机制之一。 |
| ABSTRACT： | OBJECTIVE： To detect curative mechanism of bushen huoxue huatan formula on polycystic ovarian syndrome （PCOS） model rats. METHODS： 50 rats were randomly divided into normal control group， model control group， positive control group [rosiglitazone， 3.0 mg/（kg·d）] and Bushen huoxue huatan formula high-dose， low-dose groups [7.5， 5.0 g/（kg·d）]， 10 in each group. Except for normal control group， the PCOS model was induced by Poresky method in other groups. After modeling， normal control group and model control group were received normal saline intragastrically， medicine groups were received corresponding liquids intragastrically， once a day， for 22 d. After administration， the serum levels of testosterone （T）， anti-mullerian hormone （AMH）， insulin-like growth factor Ⅰ（IGF-Ⅰ）， tumor necrosis factor-α （TNF-α） and the positive expression of transforming growth factor-β1 （TGF-β1） in ovarian tissue were determined. RESULTS： Compared with normal control group， T， AMH， IGF-Ⅰ， TNF-α levels in serum and TGF-β1 positive expression rate in ovarian tissue of rats in model control group were significantly increased （P＜0.05）； compared with model control group， T， AMH， IGF-Ⅰ， TNF-α levels in serum and TGF-β1 positive expression rate of rats in ovarian tissue in other medicine groups were significantly decreased （P＜0.05）， and the indexes in Bushen huoxue huatan formula different doses groups were superior to positive control group （P＜0.05）. CONCLUSIONS： Bushen huoxue huatan formula can obviously reduce the T， AMH， IGF-Ⅰ， TNF-α levels in serum and TGF-β1 positive expression rate in ovarian tissue in model rats with PCOS， which may be one of the mechanisms of its treatment for PCOS. |
