TITLE:
摘要: 目的:研究忧遁草枝叶的化学成分,并初步考察其体外抗肿瘤活性。方法:采用多种柱色谱技术对忧遁草95%乙醇提取物的乙酸乙酯、正丁醇部位进行分离、纯化,根据理化性质和波谱数据分析、鉴定化合物结构。采用MTT法测定鉴定出的化合物对人肝癌BEL-7402细胞、人胃癌SGC-7901细胞和人慢性髓原白血病K562细胞的体外抑制活性。结果:从忧遁草枝叶中分离鉴定出8个化合物,分别为羽扇豆醇(1)、对香豆酸甲酯(2)、1-O-acyl-3-O-(β-D-galactopyranosyl)-sn-glycerol(3)、(6R,10R)-6,10,14-trimethyl-2-pentadecanone(4)、邻苯二甲酸二(2-乙基-己基)酯(5)、α-D-甲基半乳糖苷(6)、β-D-甲基葡萄糖苷(7)、9-hydroxy-megastigma-4,7-dien-3-one-9-O-β-D-glucopyranoside(8);化合物1对人肝癌BEL-7402细胞具有弱的生长抑制作用(半数抑制浓度为42.5 μmol/L),尚未发现其他7种化合物对上述3种肿瘤细胞有抑制作用。结论:化合物2~8为首次从忧遁草枝叶中分离得到,羽扇豆醇具有弱的抗人肝癌BEL-7402细胞的活性,该研究为忧遁草的质量评价奠定了一定的基础。
ABSTRACT: OBJECTIVE: To study the chemical constituents of Clinacanthus nutans branches and leaves and its anti-tumor activity in vitro. METHODS: Varied chromatographic techniques were conducted to separate and purify the ethyl acetate and butanol parts of 95% ethanol extracts of C. nutans, the chemical structures were analyzed and identified according to physicochemical property and spectral data. MTT method was used to determine the inhibitory activity of identified compounds on human liver cancer BEL-7402 cell lines, gastric cancer SGC-7901 cell lines and chronic myelogenous leukemia K562 cell lines in vitro. RESULTS: Eight compounds isolated from C. nutans branches and leaves were identified as lupeol (1), methyl p-coumarate (2), 1-O-acyl-3- O-(β-D-galactopyranosyl)-sn-glycerol (3), (6R, 10R)-6,10,14-trimethyl-2-pentadecanone (4), 1, 2-benzenedicarboxylic acid bis (2-ethylhexyl) ester (5), methyl α-D-galactopyranoside (6), methyl β-D-glucopyranoside (7), 9-hydroxy-megastigma-4,7-dien-3-one-9-O-β-D-glucopyranoside (8). Compound 1 had weak growth inhibition effect on human liver cancer BEL-7402 cell (half inhibitory concentration was 42.5 μmol/L), the other 7 compounds had not been found to inhibit the above 3 tumor cell lines. CONCLUSIONS: Compounds 2-8 are all isolated from C. nutans branches and leaves for the first time, while lupeol has weak growth inhibition effect on human liver cancer BEL-7402 cell. The study has laid the foundation for the quality evaluation of C. nutans.
