摘要: | 目的:建立测定八角枫药材不同药用部位(茎、根茎、须根及支根)中L(-)-八角枫碱含量的方法,为八角枫药材的开发利用提供参考。方法:采用高效液相色谱法。色谱柱为Thermo C18,流动相为甲醇-磷酸缓冲液(22 ∶ 78,V/V),流速为1.0 ml/min,检测波长为259 nm,柱温为30 ℃,进样量为10 μl。结果:L(-)-八角枫碱的进样量线性范围为0.020 62~0.257 80 μg(r=0.999 9);定量限为1.7 ng,检测限为0.5 ng;精密度、稳定性、重复性试验的RSD<2%;加样回收率为97.38%~98.86%(RSD=0.6%,n=6) 。不同药用部位中L(-)-八角枫碱含量,须根>支根>根茎>茎。结论:该方法操作简便、结果准确、重复性好、专属性强,适用于测定八角枫药材不同药用部位(茎、根茎、须根及支根)中L(-)-八角枫碱的含量。 |
ABSTRACT: | OBJECTIVE: To establish a method for the content determination of L(-)-Anabasine in different medicinal parts (stem, rhizome, fibrous root, rootlet) of Alangium chinense,and provide reference for its development and utilization. METHODS: HPLC was performed on the column of Thermo C18 with mobile phase of methanol-phosphate buffer solution (22 ∶ 78,V/V) at a flow rate of 1.0 ml/min, the detection wavelength was 259 nm, column temperature was 30 ℃; the injection volume was 10 μl. RESULTS: The linear range of L(-)-Anabasine was 0.020 62-0.257 80 μg(r=0. 999 9); the limit of quantitation was 1.7 ng, limit of detection was 0.5 ng; RSDs of precision, stability and reproducibility tests were lower than 2%; recovery was 97.38%-98.86% (RSD=0.6%,n=6). The content of L(-)-Anabasine in different medicinal parts was the fibrous root>the rootlet>the rhizome>the stem. CONCLUSIONS: The method is simple, accurate, reproducible and specific, and suitable for the content determination of L(-)-Anabasine in different medicinal parts (stem, rhizome, fibrous root, rootlet) of A. Chinense. |
