摘要: 目的:比较苍术麸炒前后活性部位的药效差异。方法:将170只大鼠随机均分成17组,分别为空白对照组,湿阻脾胃模型组,生、麸炒苍术挥发油高、低剂量[分别为0.747、0.083 mg(生药)/ml,下同]组及其溶剂对照2%聚山梨酯80组,生、麸炒苍术二氯甲烷萃取物高、低剂量组及其溶剂对照1‰聚山梨酯80组,生、麸炒苍术正丁醇萃取物高、低剂量组及其溶剂对照湿阻脾胃模型蒸馏水对照组。除空白对照组外,其余16组大鼠ig番泻叶水煎液14 d建立大鼠湿阻脾胃模型,之后各组大鼠ig相应药物或溶剂,每日1次,连续7 d。测定各组大鼠给药前后体质量及给药后血清中淀粉酶、D-木糖、胃泌素、血管活性肠肽和一氧化氮(NO)的水平。结果:与湿阻脾胃模型组比较,生、麸炒苍术各活性部位组大鼠体质量增加,血清中胃泌素、淀粉酶、D-木糖水平明显升高(P<0.05),血管活性肠肽、NO水平明显降低(P<0.05);与生苍术活性部位组比较,除体质量外,麸炒苍术各活性部位组上述各指标变化更大,差异具有统计学意义(P<0.05)。结论:生苍术与麸炒苍术活性部位存在药效学差异,后者作用更强。
ABSTRACT: OBJECTIVE: To compare the pharmacodynamics of active parts in crude Atractylodes lancea and A. lancea fired with bran. METHODS: 170 rats were randomly divided into 17 groups, including blank control group, spleen and stomach damp obstruction model group, volatile oil of crude A. lancea and A. lancea fired with bran high-dose and low-dose (0.747, 0.083 mg/ml by the concentration of crude medicinal materials, similarly hereinafter) groups, solvent control 2% polysorbate 80 group, dichloromethane extract of crude A. lancea and A. lancea fired with bran high-dose and low-dose groups, solvent control 1‰ polysorbate 80 group, n-butyl alcohol extract of crude A. lancea and A. lancea fired with bran high-dose and low-dose groups, solvent control stomach damp obstruction model distilled water control group. Except blank control group, other 16 groups were given Sennae folium decoction ig for 14 d to induce spleen and stomach damp obstruction model, and then received relevant medicine or solvent ig once a day for consecutive 7 d. Body weight of rats were determined before and after medication, and the serum levels of amylase, D-xylose, gastrin, vasoactive intestinal peptide and NO were determined after medication. RESULTS: Compared with spleen and stomach damp obstruction model group, the body weight and serum levels of gastrin, amylase and D-xylose increased significantly in rats of active part in A. lancea fired with bran groups (P<0.05), while vasoactive intestinal peptide and NO decreased significantly (P<0.05). Compared active part in crude A. lancea group, except the body weight, above indicators of active part in A. lancea fired with bran groups had greater change, with statistical significance (P<0.05). CONCLUSIONS: There are pharmacodynamic differences in the active parts between crude A. lancea and A. lancea fired with bran, the latter one is stronger.
