摘要: 目的:研究三棱、莪术提取物联合热疗对结肠癌SW620细胞凋亡及迁移、侵袭能力的影响。方法:采用MTT法检测不同质量浓度的三棱提取物(1.25、2.5、5、7.5、10 μg/mL)、莪术提取物(5、10、15、20、25 μg/mL)对SW620细胞的毒性,计算其30%细胞生长抑制浓度(IC30)和50%细胞生长抑制浓度(IC50)。采用CFSE/PI双荧光染色法、Annexin V/PI双染色法+流式细胞术考察IC50三棱提取物、IC50莪术提取物分别单用或联合热疗(42 ℃条件下处理90 min)对SW620细胞死亡情况和凋亡率的影响;采用划痕实验和Transwell侵袭实验考察IC30三棱提取物、IC30莪术提取物分别单用或联合热疗对SW620细胞迁移和侵袭能力的影响。结果:三棱提取物IC30为3.24 μg/mL,IC50为4.69 μg/mL;莪术提取物IC30为11.27 μg/mL,IC50为16.81 μg/mL。与IC50三棱提取物组或IC50莪术提取物组比较,IC50三棱提取物+热疗组或IC50莪术提取物+热疗组的死亡细胞显著增多,细胞凋亡率显著升高(P<0.05)。与0 h时比较,IC30三棱提取物组、IC30莪术提取物组、IC30三棱提取物+热疗组、IC30莪术提取物+热疗组细胞划痕实验的愈合间距均未见明显变窄。与IC30三棱提取物组或IC30莪术提取物组比较,IC30三棱提取物+热疗组或IC30莪术提取物+热疗组侵袭实验的跨膜细胞数均显著减少(P<0.05)。结论:联用热疗能显著增强三棱或莪术提取物对SW620细胞的杀伤作用,并能显著提高两种提取物对SW620细胞迁移和侵袭能力的抑制作用。
ABSTRACT: OBJECTIVE: To study the effects of Sparganium stoloni extract or Curcuma zedoaria extract combined with hyperthermia treatment on apoptosis, migration and invasion ability of colon cancer SW620 cells. METHODS: MTT assay was performed to detect the toxicity of S. stoloni extract (1.25, 2.5, 5, 7.5, 10 μg/mL) and C. zedoaria extract (5, 10, 15, 20, 25 μg/mL) to SW620 cells. The corresponding IC30 and IC50 were calculated. CFSE/PI double-staining method and Annexin V/PI double-staining+flow cytometry were used to investigate the effects of IC50 S. stoloni extract or IC50 C. zedoaria extract combined with hyperthermia treatment on the death and apoptotic rate of SW620 cells. Scratch test and Transwell invasion test were used to investigate the effects of IC30 S. stoloni extract or IC30 C. zedoaria extract combined with hyperthermia treatment on the migration and invasion ability of SW620 cells. RESULTS: IC30 and IC50 of S. stoloni extract were 3.24 μg/mL and 4.69 μg/mL; those of C. zedoaria extract were 11.27 μg/mL and 16.81 μg/mL. Compared with IC50 S. stoloni extract group or IC50 C. zedoaria extract group, death cells and apoptotic rate were increased significantly in IC50 S. stoloni extract+hyperthermia treatment group or IC50 C. zedoaria extract+hyperthermia treatment group (P<0.05). Compared with 0 h, healing distance of scratch test was not significantly narrowed in IC30 S. stoloni extract group, IC30 C. zedoaria extract group, IC30 S. stoloni extract+hyperthermia treatment group or IC30 C. zedoaria extract+hyperthermia treatment group. Compared with IC30 S. stoloni extract group or IC30 C. zedoaria extract group,  the number of transmembrane cells decreased significantly in the invasive experiment in IC30 S. stoloni extract+hyperthermia treatment group or IC30 C. zedoaria extract+hyperthermia treatment group (P<0.05). CONCLUSIONS: Hyperthermia combination can significantly enhance the killing effect of S. stoloni extract or C. zedoaria extract on SW620 cells, and significantly improve their inhibitory effects on SW620 cell migration and invasion.
