摘要: | 目的:研究苗药理气活血滴丸对慢性心力衰竭(CHF)模型大鼠的保护作用。方法:将60只SD大鼠随机分为正常组(10只)和造模组(50只),造模组大鼠皮下注射异丙肾上腺素(ISO)以复制CHF模型。将造模成功的大鼠随机分为模型组、依那普利组(阳性对照,20 mg/kg)和理气活血滴丸低、中、高剂量组(43.75、87.50、175.00 mg/kg),每组10只。各给药组大鼠每天灌胃相应药物1次,正常组和模型组大鼠均灌胃等容生理盐水。连续给药4周后,监测大鼠心功能指标[左心室内压峰值(LVSP)、左心室舒张末压(LVEDP)、左心室内压最大上升速率(+dp/dtmax)、左心室内压最大下降速率(-dp/dtmax)];计算左心质量指数;采用苏木精-伊红染色观察心肌组织形态学变化;采用Masson染色观察心肌纤维化情况,并计算胶原容积分数(CVF);采用酶联免疫吸附测定法检测血清中脑钠肽(BNP)、肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)的含量。结果:与正常组比较,模型组大鼠心肌组织损伤严重,心肌纤维化程度明显加重,其LVSP、±dp/dtmax均显著降低,LVEDP、左心质量指数、CVF以及血清中BNP、TNF-α、IL-6的含量均显著升高(P<0.05或P<0.01)。与模型组比较,依那普利组和理气活血滴丸中、高剂量组大鼠心肌组织损伤和心肌纤维化程度均明显减轻,其LVSP、±dp/dtmax均显著升高,LVEDP、左心质量指数、CVF以及血清中BNP、TNF-α、IL-6的含量均显著降低(P<0.05或P<0.01)。结论:理气活血滴丸对ISO诱导的CHF模型大鼠具有一定的保护作用,能显著改善其心功能指标,减轻心肌组织受损和心肌纤维化程度,降低血清中BNP、TNF-α、IL-6的含量。 |
ABSTRACT: | OBJECTIVE: To study the protective effects of Miao medicine Liqi huoxue dropping pills on chronic heart failure (CHF) model rats. METHODS: Totally 60 SD rats were randomly divided into normal group (10 rats) and modeling group (50 rats). Modeling group was given subcutaneous injection of isoproterenol (ISO) to induce CHF model. After modeling, CHF model rats were randomly divided into model group, enalapril group (positive control, 20 mg/kg) and Liqi huoxue dripping pills low-dose, medium-dose and high-dose groups (43.75, 87.50, 175.00 mg/kg), with 10 rats in each group. Administration groups were given relevant medicine intragastrically once a day; normal group and modeling group were both given constant volume of normal saline intragastrically. After 4 weeks of administration, the cardiac function indexes (LVSP, LVEDP, +dp/dtmax, -dp/dtmax) of rats were monitored. The left ventricular mass index was calculated. The morphologic changes of myocardial tissue were observed by HE staining. The myocardial fibrosis was observed by Masson staining, and CVF was calculated. The serum contents of BNP, TNF-α and IL-6 were detected by ELISA. RESULTS: Compared with normal group, there was severe myocardial tissue injury of rats and the degree of myocardial fibrosis increased significantly in model group; the levels of LVSP and ±dp/dtmax were decreased significantly, while LVEDP, left ventricular mass index, CVF, serum contents of BNP, TNF-α and IL-6 were increased significantly (P<0.05 or P<0.01). Compared with model group, myocardial tissue injury and the degree of myocardial fibrosis were relieved significantly in enalapril group and Liqi huoxue dripping pills medium-dose and high-dose groups; while the levels of LVSP and ±dp/dtmax were increased significantly; LVEDP, left ventricular mass index, CVF, serum contents of BNP, TNF-α and IL-6 were decreased significantly (P<0.05 or P<0.01). CONCLUSIONS: Liqi huoxue dripping pills can protect ISO-induced CHF model rats, improve cardiac function indexes, relieve myocardial tissue injury and the degree of myocardial fibrosis and reduce the serum contents of BNP, TNF-α and IL-6. |
