摘要: 目的:建立小儿柴桂退热颗粒的超高效液相色谱(UPLC)指纹图谱,并进行聚类分析与主成分分析。方法:采用UPLC法,色谱柱为 Waters Acquity UPLC BEH C18,流动相为甲醇-0.05%磷酸溶液(梯度洗脱),流速为0.5 mL/min,检测波长为260 nm,柱温为40 ℃,进样量为0.5 μL。以葛根素为参照,测定14批小儿柴桂退热颗粒样品的UPLC图谱,采用《中药色谱指纹图谱相似度评价系统》(2012版)对14批样品进行相似度评价,确定共有峰。采用SPSS 19.0软件进行聚类分析与主成分分析。结果:14批样品的UPLC图谱有15个共有峰,相似度均>0.90;经验证,14批样品UPLC图谱与对照指纹图谱具有较好的一致性。14批样品可聚为两大类。结论:所建指纹图谱可为小儿柴桂退热颗粒的质量评价提供参考。
ABSTRACT: OBJECTIVE: To establish UPLC fingerprint of Xiao’er chaigui tuire granules, and to conduct cluster analysis (CA) and principal component analysis (PCA). METHODS: UPLC method was adopted. The determination was performed on Waters Acquity UPLC BEH C18 column with mobile phase consisted of methanol-0.05% formic acid solution (gradient elution) at the flow rate of 0.5 mL/min. The detection wavelength was 260 nm, the column temperature was 40 ℃, and sample size was 0.5 μL. Using puerarin as reference, UPLC fingerprints of 14 batches of Xiao’er chaigui tuire granules were determined. The similarity of 14 batches of samples was evaluated by using TCM Chromatographic Fingerprint Similarity Evaluation System (2012 edition), and common peak was identified. CA and PCA were conducted by using SPSS 19.0 software. RESULTS: There were 15 common peaks in UPLC chromatograms of 14 batches of samples. The similarity of UPLC fingerprints of 14 batches of samples was higher than 0.90, and UPLC fingerprint was in good agreement with control fingerprint. The 14 batches of samples could be clustered into 2 categories. CONCLUSIONS: Established fingerprint can provide reference for quality evaluation of Xiao’er chaigui tuire granules.
