摘要: | 目的:优化头孢噻肟钠原料药有关物质检查中溶液制备条件。方法:采用高效液相色谱法检测头孢噻肟钠原料药杂质总量;以磷酸盐缓冲液pH、放置温度、放置时间和光照条件为因素,以杂质总量为指标,在单因素试验基础上,采用L9(34)正交试验优化溶液制备条件,并进行验证试验。结果:优化的溶液制备方法为避光操作,磷酸盐缓冲液的pH为6.50,试验环境温度和控制样品温度为5 ℃,制备溶液后5 min内进样。结论:优化后的方法重复性好、结果可靠。 |
ABSTRACT: | OBJECTIVE: To optimize the solution preparation in related substance test of cefotaxime sodium. METHODS: HPLC method was adopted to determine the total amount of impurities in cefotaxime sodium. Using phosphoric acid buffer solution pH, placing temperature, standing time and illumination as factor, the total amount of impurities as indexes, the preparation condition of solution was optimized by L9(34) orthogonal test. The validation test was carried out. RESULTS: The optimal preparation method was as follows as the sample was dissolved with a solvent containing pH 6.50 phosphate buffer protected from light. The test temperature and the sample temperature were set at 5 ℃, and the sample was injected within 5 min after the preparation of the solution. CONCLUSIONS: The optimized method is reproducible and reliable. |
