摘要: | 目的:比较黄芪破壁饮片煎煮、冲泡与传统饮片煎煮后有效成分溶出含量的差异,为黄芪破壁饮片临床应用提供参考。方法:通过正交试验确定黄芪破壁饮片和传统饮片最优的煎煮工艺[液料比(mL/g)、煎煮次数、煎煮前浸泡时间(min)]以及破壁饮片最优冲泡工艺[水温(℃)、液料比、冲泡次数],在各自的最优工艺下比较5种成分(黄芪甲苷、毛蕊异黄酮葡萄糖苷、芒柄花苷、毛蕊异黄酮、芒柄花素)在不同煎煮/冲泡时间点的溶出含量差异。结果:黄芪破壁饮片按最优煎煮工艺(液料比50 ∶ 1,煎煮前不浸泡,煎煮2次)煎煮5~10 min时其有效成分溶出含量已达到传统饮片煎煮(煎煮前浸泡30 min,液料比50 ∶ 1,煎煮3次)30 min的溶出含量;其在最优冲泡工艺(液料比75 ∶ 1,水温100 ℃,冲泡3次)下有效成分可以达到传统饮片煎煮时溶出含量的80%~90%。结论:黄芪饮片经过破壁后有效成分溶出速率更快,可以免去煎煮前浸泡时间,并减少煎煮时间、次数,煎煮效率显著提高;临床使用时可采用冲泡方式。 |
ABSTRACT: | OBJECTIVE: To compare the dissolution content differences of active ingredients in Astragali radix cell-broken piece after decocting and socking and traditional piece after decocting, and provide reference for the clinical use of Astragali radix cell-broken piece. METHODS: Orthogonal test was used to ensure the optimal decoction technology [solid-liquid ratio (mL/g), decoction times, socking time before decoction (min)] and socking technology [water temperature (℃), liquid-solid ratio, socking times] of Astragali radix cell-broken piece and traditional piece. The dissolution content differences of astragaloside A, calycosin-7-glucoside, ononin, calycosin, formononetin at different decoction and socking time points were compared under their optimal technology. RESULTS: The dissolution contents of active ingredients in Astragali radix cell-broken piece under the optimal decoction technology (liquid-solid ratio of 50 ∶ 1, no socking before decoction, decocting twice) when decocting for 5-10 min were equal to the contents of traditional piece when decocting for 30 min (socking 30 min before decoction, liquid-solid ratio of 50 ∶ 1, decocting 3 times). The active ingredient can reach the 80%-90% of dissolution contents of traditional piece under optimal socking technology (liquid-solid ratio for 75 ∶ 1, water temperature of 100 ℃, socking 3 times). CONCLUSIONS: After cell-broken, the dissolution speed of active ingredients was faster, Astragali radix piece can avoid the socking time before decoction, reduce decoction time and times, significantly improve decoction efficiency. It can be used with soaking in clinical. |
