摘要: 目的:采用微流控芯片非接触电导法测定异烟肼片中异烟肼的含量。方法:探讨缓冲溶液的种类、浓度、pH、添加剂种类和添加量、进样时间、分离电压等因素对分离检测的影响。结果:优选缓冲体系为1.86 mmol/L柠檬酸-0.14 mmol/L柠檬酸钠缓冲溶液(pH3.0),不加添加剂,进样时间为20 s,分离电压为2.5 kV。优化条件下,异烟肼检测质量浓度线性范围为10~200 μg/mL(r=0.999 3);定量限为8.0 μg/mL,检测限为2.4 μg/mL;精密度、稳定性、重复性试验的RSD<2.0%;加样回收率为97.8%~102.9%(RSD=1.9%,n=9)。结论:该方法快速简便,适用于测定异烟肼片中异烟肼的含量。
ABSTRACT: OBJECTIVE: To establish a method for the content determination of isoniazid in Isoniazid tablet. METHODS: Microfluidic chip with contactless conductivity detection was adopted. The effects of electrophoresis parameters, such as the variety, concentration, pH, additive types and dosage, injection time and separation voltage on the separation detection were explored. RESULTS: The optimized buffer system was 1.86 mmol/L citrate-0.14 mmol/L sodium citrate buffer solution (pH3.0),no additive,the injection time was 20 s and the separation voltage was 2.5 kV. In the optimal conditions, the linear range of isoniazid was 10-200 μg/mL(r=0.999 3); the limits of quantification and detection were 8.0 μg/mL and 2.4 μg/mL; RSDs of precision, stability and reproducibility tests were lower than 2.0%; recovery was 97.8%-102.9% (RSD=1.9%,n=9). CONCLUSIONS: The method is rapid and simple, and suitable for the content determination of isoniazid in Isoniazid tablet.
